چکیده

Orexins are neuropeptides that play important roles in sleep-wake regulation and food intake the central nervous system, but their receptors also expressed peripheral tissues, including endocrine system. In present study, we investigated functions of orexin adrenal steroidogenesis using human adrenocortical H295R cells by focusing on its interaction with bone morphogenetic proteins (BMPs) induce steroidogenesis. Treatment A increased mRNA levels steroidogenic enzymes StAR, CYP11B2, CYP17, HSD3B1, these effects were further enhanced presence forskolin. Interestingly, treatment suppressed BMP-receptor signaling detected Smad1/5/9 phosphorylation Id-1 expression through upregulation inhibitory Smad7. Orexin endogenous BMP-6 type-II receptor ActRII cells. Moreover, downregulated level OX1R, not OX2R, conclusion, results indicate both accelerate cells; pathways mutually inhibit each other, thereby leading to a fine-tuning

Bone morphogenetic proteins 2 and 5 are down-regulated in adrenocortical carcinoma and modulate adrenal cell proliferation and steroidogenesis.

Bone morphogenetic proteins (BMP) have been shown to affect tumorigenesis in a variety of tumors. Quantitative PCR analysis revealed down-regulation of BMP2 and BMP5 in tissue samples from adrenocortical carcinoma and adrenocortical tumor cell lines compared with normal adrenal glands. Integrity of BMP-dependent pathways in these cell lines could be shown by activation of the Smad1/5/8 pathway ...

Bone morphogenetic proteins in human bone regeneration.

Recently, the first clinical reports on bone regeneration by two recombinant human bone morphogenetic proteins (rhBMPs), BMP-2 and BMP-7 (also named osteogenic protein-1, OP-1) have been published (1-4). Although both BMPs were able to support bone regeneration, a significant variation in individual response was observed with both proteins. Animal studies and laboratory experiments reveal a num...

Bone morphogenetic proteins differentially regulate pigmentation in human skin cells.

Bone morphogenetic proteins (BMPs) are a large family of multi-functional secreted signalling molecules. Previously BMP2/4 were shown to inhibit skin pigmentation by downregulating tyrosinase expression and activity in epidermal melanocytes. However, a possible role for other BMP family members and their antagonists in melanogenesis has not yet been explored. In this study we show that BMP4 and...

Regeneration of the skeleton by recombinant human bone morphogenetic proteins.

Recombinant human bone morphogenetic proteins (rhBMPs) have past a long journey in human orthopaedic surgery during the last 15 years. From the first reports of the use of rhBMPs in hostile environments such as critically-sized bone defects, avascular femoral head necrosis, unstable thoracolumbar vertebral fractures, instability between the atlas and axis due to rheumatoid arthritis; over the u...

Roles of bone morphogenetic protein-6 in aldosterone regulation by adrenocortical cells.

Aldosterone production occurs in the adrenal cortex, and is regulated primarily by angiotensin II (Ang II), potassium and adrenocorticotropin (ACTH). In the presence of the aldosterone stimulators, steroidogenesis is further governed by local autocrine and/or paracrine factors in the adrenal cortex. We reported the presence of functional bone morphogenetic protein (BMP) system in the adrenal co...
